For a resale flat specifically, a valid HFE letter is required before HDB will allow the buyer to receive the Option to Purchase, not after, so applying early in the search process avoids delays once a flat is found.
The letter is valid for 9 months and HDB does not extend it once it expires, a fresh application is required if a purchase has not completed within that window, so timing the application against the expected pace of the property search matters.
